" Last CSH meeting in San Mateo, Mary Daniel from Diagnostic BioSystems told me to try out their EDTA Buffer since I was having problems with stabilizing the pH of the EDTA that we were using in the laboratory. The buffer comes in 500mL of 10X concentration. It is extremely convenient that we can just make up a bottle of working dilution of EDTA and use it until it was all gone and do not have to worry about the change in the pH after a day or so. Since we are happy with the perfomance of this product. "

" I was wondering if anyone had been working with or had any knowledge of a couple of products from Diagnostic Biosystems. They are the Histozyme retrieval system and the Intensi/fire DAB enhancement system. I've been using the Histozyme in place of both pepsin and trypsin for 5 minutes at room temperature. The preliminary results have been outstanding. I'ts a big time saver. The enhancement solution has allowed me to use my antibodies at one tenth of the dilutions I normally use. This week I'm in the process of diluting them even further to see what the results are. I'm hoping it will allow me to stop using edta as much as possible and only use citrate retrieval. If anyone has any experience with either of these products please advise me as to what you are doing and how they are working. "

"We were very happy to see the results of our tests with Histo/Zyme in IHC. We obtained manifold higher signal intensities for several Cytokeratin stains with 5 minutes Histo/Zyme at room temperature compared to 10 minutes Pronase at 37 or to 10 minutes Trypsin at 37, respectively. Even more stunning were our results with certain Collagen antibodies. We obtained the same staining intensity with 75 minutes Pepsin at 37 and with 5 minutes Histo/Zyme at room temperature. Compared to Pepsin pre treatment with Histo/Zyme resulted in significantly better cellular morphology."
